In biology we shouldn't be aiming to reproduce exact experiments but to find the same conclusions and observations with different approaches - that's how you can be confident in a finding. Now if an entire paper (or more importantly it's conclusions) can't be reproduced then yes that is a problem. If details to replicate experiments are not provided then that too is a problem. For example, cell culture density can have huge effects on signalling pathways and yet it's not something that is monitored as closely or reproducibly as it should be. Room temperature can also have a big effect (or how quickly you work) yet how many are recording this daily?
