We find the cells get pretty stressed out after about 20-30 days, so we usually add serum when we have lengthy protocols that involve transfections and clone picking and other stressors.

Mouse ESC people: we are looking for serum replacement options largely driven by ethical reasons. This is mainly for keeping cells happy when we make transgenics/mutants as we usually go serum free for experiments. Any good advice out there? Comments welcome!
