Thank you! Regarding the deep sequencing: Yes, I fully agree that's the next step. This is something that we are looking at right now to get an overview which host factors this phage (and others) need under different conditions.

I am trying the optimized TSS procedure of @ATinyGreenCell.bsky.social that doesn't require to first grow the bugs to exponential phase. Would be great if this works in our hands (at least with E.coli K-12 strains)!

While it sounds superficially similar to relevant bugs, this strain lacks proper DNA repair, stringent response, and O-antigen... so physiological data generated with it might not be what you want your next treatment to be based on.
