🧬🧶 Pre-print out now: 🌲 S-lignin O-demethylation by a heme peroxygenase. If you're lucky enough to have a student like Alix Harlington, you'll know the warm glow you get from supporting work of this calibre. Check out our current favourite #P450tinyurl.com/nfe36afb

🧬🧶 #lorneproteins ⚡️ Gabby Watson - Nbs to help cross the BBB. E.g. Transferrin receptor - repurpose? Brain shuttle. Fab+Ab that targets Tfr1. 940 Nbs screened 🦸‍♀️. Binding affinity- high affinity isn't good. Likely to get routed to lysosomes. Mod affinity. Fluo label - 🧠✅️🫶👏

🧬🧶 #lorneproteins ⚡️ Joanna New: Tauopathies - symptoms of CTE overlap with other Tauopathies. Only evident in death. Characterising novel amyloid binding probes. Recombinant Tau fibrils - observe probe+/- amyloid. 👏🫶

🧬🧶 #lorneproteins ⚡️ @acmarshallphd liquid protein demixing for subcellular organisation. DBHS proteins RNA/DNA binding proteins. In paraspeckles. Prion-like N-LCR not main driver of demixing... attenuated. C-LCR higher pass separation. 🫶👏 Great talk Andrew. 🎷👌🤣

#lorneproteins ⚡️ talks: Vanessa Morris-Caspase8 master reg apoptosis. Dimerisation, autocat cleavage to active dimer. HOSCN impact on cell death pathways? Necroptosis...unpublished? Maybe. 🤐🤩👏

Prolonged stress response signaling but not protein aggregation causes cell death. 🤯Treatment? Pharmacological intervention for stress response silencing 🫶👏👏👏 Amazing talk!

Control stability and localisation- SIFI degron. Stress response silencing is an active process. SIFI detects misfolded mislocalised mit proteins. Then targets the stress signaling and shuts that down. Stress response silencing?

When does this happen in cells? Atf4 TF readout of cell stress. DeltaUbr4 - cell has a lot of trouble cleaning up after stress. Signals need to be actively turned off. Timing? Amphipathic helices are degrons that interact with Ubr4. Mitochondrial pre-sequences also!

HRI and DELE1. Dele1/HRI signals to shut down translation when mit import fails and aggregates are accumulating. SIFI complex only targets active DELE1/HRI for ubiuqitylation. Ubr ko active DELE1 stable.
