We're charging ahead on this work, modeling the SNP in ovarian/gyn contexts in vitro and in vivo to learn more about how diff variant allele freq corresponds to diff gyn cancer biology and poor outcomes! Need to write the grants for the lobular half of the story now... 😅

5) The SNP is common, but variable - ranges from 0% in Black populations up to >50% in East Asian populations, and everywhere in between. 6) SNP may link gyn pathology risk (incr risk with variant allele) to outcomes disparities (diff tumor biology in our work)

4) WNT4 gene 'activation' is via converging mechanisms - in lobular, hijacked by estrogen receptor; in ovarian/gyn, a common SNP (at the lobular ER binding site) confers new regulatory context and activation.

1) Rather than typical Wnt secretion, we see WNT4 lives at mitochondria. Supports our prior data re: non-palmitoylated intracellular WNT4. 2) WNT4 controls shift from glucose metab --> more fatty acid/amino acid metabolism. 3) We see #2 in ILC+OvCa cell lines, and in Gyn tumors
