Some people like protein spikes but I’m lazy. Could digest a “standard” like a pool from Golden West or NIST. We also make a study-specific pool and optimally digest that multiple times.

This discussion reminds me of my PhD preliminary assignment from those HUPO Human Plasma Proteome Project days.. I had to summarise a whole special issue of the journal Proteomics for that assignment..

Echoing @mattwfoster.bsky.social but we use NIST 909c or 1950 as an EQC, but I don’t know how helpful it is other than a sanity check on digestion and processing. As Matt says, also have an SPQC and I’d argue some other external peptide mix (hela or Hek) to monitor the run independent of method.
